Laser Radiation Mutagenesis Used in Thermostability Phytase Screening

Abstract: Phytase producing Aspergillus Niger was treated with 1.06¦Ìm,0.53¦Ìm and 1.06+0.53¦Ìm high repetition rate Nd:YAG laser by sequential and pulse radiate manner. From the result of mutagenesis and compare the product of the forward mutate ratio and the forward mutate range, it showed that the mutate result of sequential better than pulse's; Among the sequential manner, the mutate effect of 1.06¦Ìm laser radiation was prominence with 15W power and 1 min radiate time, to 0.53¦Ìm laser, it should protract the radiate time; Among the pulse radiate manner, mutate effect of 1.06¦Ìm laser radiation was better and the suitable pulse number was 100~200.One mutate fungus that produced thermostability phytase was screened, the effect of high temperature was lower, its activity remain 44% after keep 10min under 80¡æ condition.
