Abstract: The developmental morphology of plant cell was studied in leaves and roots of Arabidopsis thaliana in this paper. The surface of lamina in Arabidopsis with the different size was scanned by UV laser(351 nm), the adaxial epidermis cells of lamina were clearly divided into five distinct cell-types, i.e. epidermal pavement cells with different size, stomata, subsidiary cells, trichomes (hair-like cells) and socket cells in the leaves with differentiation. The root tip of Arabidopsis was scanned by blue laser£¨488 nm£©with light-staining by eosin, different kinds of cells, such as epidermis cell, cortex cell, quiescent cells, cortex/endodermal initial cell, epidermal/root cap could easily be distinguished in the root tip. Autofluoresence from chloroplast was seen in the leaves of Arabidopsis after the leaves were scanned continuously by multiphotone£¨800 nm£© for 30 seconds, and the trace of movement of chloroplast was clearly observed in mesophyll cell of leaves. It showed that CLSM was a very novel tool to study the morphology and structure of plant cell .
