Objective: To investigate the mechanism of a novel amphipathic photo sensitizer [di-sulfonated di-phthalimidomethyl phthalocyanine zinc (ZnPcS2P2)]-mediated photodynamic therapy (ZnPcS2P2-PDT) on K562 cells and HL60 cells.Methods: Laser scanning confocal microscopy was applied and three special organelle probes including Rhodamine123, Lys -oTracker DND-26 and Dioc6(3) were selected to label mitochondria,lysosome and endoplasmic reticulum fluorescence images both of ZnPc- S2P2 and the three probes were collected through corresponding emission filters. Results: ZnPcS2P2 located on mitochondria lysosome. Endoplasmic reticulum.Conclusion: this study provides direct evidence that mitochondria£¬lysosome and endoplasmic reticulum play an important role in the apoptosis in K562 cells and HL60 cells induced by ZnPcS2P2 - PDT.
