稀释液中添加海藻糖对山羊精子功能和膜完整性的影响

摘 要:选用5只年龄为3-4岁的波尔山羊公羊研究在稀释液中添加海藻糖对山羊精子功能和膜完整性的影响。山羊精子分别用含6.6、13.2、19.8、26.4、39.6、52.9、66.1、79.3 mmol/L的不同海藻糖的Tris-柠檬酸-葡糖糖(TCG)稀释液(卵黄:18 %;甘油:6 %)稀释和冷冻。结果表明,39.6、52.9、66.1、79.3 mmol/L组降温后的精子活率显著(P<0.05)降低;52.9、66.1、79.3 mmol/L组降温后的精子畸形率和39.6 mmol/L组降温后的膨胀精子率显著(P<0.05)提高。26.4 mmol/L组和39.6 mmol/L组冻融后的精子活率显著(P<0.05)高于对照组;66.1 mmol/L和79.3 mmol/L组冻融后的精子活率、畸形率分别显著(P<0.05)低于和高于对照组。19.8、26.4、39.6mmol/L组冻融后精子获能率显著(P<0.05)低于对照组。39.6 mmol/L组冻融后顶体完整率和膨胀精子率显著(P<0.05)高于对照组,而66.1和79.3 mmol/L组显著(P<0.05)低于对照组。39.6 mmol/L组的受胎率显著(P<0.05)高于对照组,而66.1mmol/L组和79.3 mmol/L组的受胎率显著(P<0.05)低于对照组。表明,在含18%的卵黄(v/v)、6%甘油(v/v)的TCG稀释液中,添加适宜浓度(26.4 mmol/L和39.6 mmol/L)海藻糖,可显著提高山羊精子功能和膜的完整性。


Effects of Trehalose in Semen Diluents on Function and Membrane Integrity of Goats Spermatozoa

Abstract: Five male boar goats with 3-4 years old were selected to study the influence of trehalose in Semen Diluents on function and membrane integrity of goat spermatozoa. Goat spermatozoa were diluted(1:4) and frozen in Tris-citric acid-glucose (TCG) diluents with different concentrations of trehalose(0, 6.6, 13.2, 19.8, 26.4, 39.6, 52.9, 66.1, 79.3 mmol/L) respectively. Final concentrations of 18% (v/v) egg yolk and 6% (v/v) glycerol were employed in the diluents. The results showed that, compared with control(0 mmol/L ), sperm motilities of 39.6, 52.9, 66.1, 79.3 mmol/L groups were decreased significantly(P<0.05), abnormal sperm rates of 52.9, 66.1, 79.3 mmol/L groups and swollen sperm rate of 39.6 mmol/L group were increased significantly(P<0.05) after cooling (P<0.05). Compared with control, sperm motilities of 24.6 mmol/L and 39.6mmol/L groups were increased significantly(P<0.05), sperm motility, abnormal sperm rate of 66.1 mmol/L and 79.3 mmol/L groups were decreased and increased significantly(P<0.05) after freezing and thawing respectively. Frozen-thawed sperm capacitation rates that developed from sperm with trehalose were 19.8, 26.4 and 39.6 mmol/L, which were significantly(P<0.05) higher than sperm without trehalose. Frozen-thawed intact acrosomes rate and swollen sperm rate of 39.6 mmol/L trehalose group were significantly(P<0.05) higher than control , but their rates of 66.1 mmol/L, 79.3 mmol/L trehalose groups were decreased significantly(P<0.05) than control proup. Compared with control, frozen-thawed conception rate of 39.6 mmol/L group was increased significantly(P<0.05), but its rate of 66.1 mmol/L and 79.3 mmol/L groups were decreased significantly(P<0.05). The results showed that goat sperm function and membrane integrity were significantly(P<0.05) improved, while adding suitable concentration(26.4 mmol/L or 39.6 mmol/L) trehalose in the TCG diluents with 18% (v/v) egg yolk and 6% (v/v) glycerol.
